      技術(shù)領(lǐng)域
      [0001] 本發(fā)明屬于微生物技術(shù)領(lǐng)域,具體涉及一種高轉(zhuǎn)化率的枯草芽孢桿菌及其制備與 應(yīng)用。
      【背景技術(shù)】
      [0002] 枯草芽孢桿菌被認(rèn)定為是一般安全(GRAS,Generally Recognized as Safe)的生 物,因其無(wú)致熱源、高分泌、易培養(yǎng),其產(chǎn)物可直接用于人們的日常使用等優(yōu)點(diǎn),成為工業(yè)多 肽類產(chǎn)品、藥劑和酶制劑分泌表達(dá)的首選??莶菅挎邨U菌A164(ATCC 6051a)相比于模式菌 株A168,其生長(zhǎng)特性、分泌能力等生化特征非常優(yōu)異,是發(fā)酵工業(yè)生產(chǎn)菌構(gòu)建的理想出發(fā)菌 株。但是A164由于DNA轉(zhuǎn)化效率極抵,基因操作困難,使得A164工程菌改造的工作限制大,影 響了其在工業(yè)生產(chǎn)應(yīng)用的前景。
      [0003] 枯草芽孢桿菌的攝取外源DNA的能力,即感受態(tài)效率受到信號(hào)轉(zhuǎn)導(dǎo)網(wǎng)絡(luò)嚴(yán)密的調(diào) 控。感受態(tài)轉(zhuǎn)錄因子Comk在其中扮演著重要的作用,Comk的編碼基因 comk的表達(dá)也在轉(zhuǎn)錄 和轉(zhuǎn)錄后水平上受到嚴(yán)格調(diào)控。comk的表達(dá)受到至少三個(gè)轉(zhuǎn)錄因子,包括AbrB、Rok(Ykuw) 和CodY的抑制,同時(shí)也受到DeqSu雙組分調(diào)控系統(tǒng)及Comk本身的正向調(diào)控。因此,提高枯草 芽孢桿菌的感受態(tài)效率,可以從兩個(gè)方面著手,其一是敲除comk基因的抑制因子,其二就是 過(guò)表達(dá)Comk正向調(diào)控因子。事實(shí)證明,這兩種方式均切實(shí)可行,通過(guò)失活Rok的編碼基因 rok,可以使枯草芽孢桿菌BD630的轉(zhuǎn)化效率提高5倍;而通過(guò)將含有木糖啟動(dòng)子xylA的comk 基因?qū)肟莶菅挎邨U菌1A751,得到的枯草芽孢桿菌1A976對(duì)DNA的轉(zhuǎn)化效率可以提高1000 倍。
      [0004] 另一方面,經(jīng)基因工程改造的微生物通常含有抗生素基因以用于遺傳操作篩選, 但用于多肽、工業(yè)酶生產(chǎn)的領(lǐng)域?qū)股氐奶砑蛹翱股鼗虻拇嬖谑置舾?,因此,一?要求生產(chǎn)菌株不含抗生素基因。因此,在對(duì)枯草芽孢桿菌進(jìn)行分子改造時(shí),通常會(huì)使用反向 篩選標(biāo)記,利用細(xì)胞自身的同源重組系統(tǒng)將抗性基因敲除,但這種方式下的重組效率不高, 菌株二次篩選時(shí)的工作量比較大,也比較耗時(shí);因此近年來(lái)多利用位點(diǎn)特異性重組系統(tǒng),比 如cre/lox系統(tǒng),通過(guò)外源重組酶的催化,大大提高重組效率,使得基因無(wú)痕敲除/敲入的操 作省時(shí)高效。
      [0005] 但是即使是利用現(xiàn)代的遺傳操作工具,雖然它們對(duì)枯草芽孢桿菌A168、1A976等模 式菌株十分有效,但是對(duì)于有生產(chǎn)應(yīng)用前景的A164而言,依舊十分困難,因?yàn)锳164的轉(zhuǎn)化效 率極低,幾乎不接受外源DNA的轉(zhuǎn)化。同時(shí),為了保證外源基因在枯草芽孢桿菌的遺傳穩(wěn)定 性,會(huì)通過(guò)同源重組整合到宿主的染色體中,利用線性DNA往往可以一次性完成這種分子改 造,但對(duì)于A164而言,這種由線性DNA介導(dǎo)的分子改造工作十分困難,耗時(shí)耗力。

      【發(fā)明內(nèi)容】

      [0006] 為了克服目前野生型枯草芽孢桿菌轉(zhuǎn)化效率低的問(wèn)題,本發(fā)明的目的在于提供一 種高轉(zhuǎn)化率的枯草芽孢桿菌及其制備與應(yīng)用。
      [0007] 為了實(shí)現(xiàn)上述目的以及其他相關(guān)目的,本發(fā)明采用如下技術(shù)方案:
      [0008] 本發(fā)明的第一方面,提供了一種枯草芽孢桿菌,所述枯草芽孢桿菌的基因組中含 有xylR-Pxy lA-comk表達(dá)單元,所述xylR-PxylA-comk表達(dá)單元的序列包括5 '至3 '方向依次 排列的xylR基因序列、xylA啟動(dòng)子基因序列、comk基因序列。

      [0108] 采用誘導(dǎo)轉(zhuǎn)化的方式,可以方便地對(duì)枯草芽孢桿菌164S進(jìn)行基因操作。利用限制 性內(nèi)切酶Kpn I切割淀粉酶敲除質(zhì)粒pDG364后,不做產(chǎn)物的清潔,直接轉(zhuǎn)化枯草芽孢桿菌 164S:
      [0109] a)把平板活化的枯草芽孢桿菌164S落接種到3mL LB試管中,37°C、200rpm培養(yǎng)10 ~12h;
      [0110] b)然取300yL培養(yǎng)液轉(zhuǎn)接到預(yù)熱的3mL LB試管中,加入濾膜滅菌的的木糖溶液至 木糖終濃度為1%,37°C、200rpm培養(yǎng)3h;
      [0111] c)分裝到2mL Eppendorf管中,按比例IOOng DNA/100yL培養(yǎng)液加入線性化的質(zhì)粒 后孵育,37 °C、200rpm培養(yǎng)1.5h。涂5yg/mL的氯霉素抗性板。37 °C,過(guò)夜培養(yǎng)。
      [0112]挑取單克隆轉(zhuǎn)接篩選得到的菌于含2%。(質(zhì)量分?jǐn)?shù))可溶性淀粉的LB平板上,37°C 過(guò)夜培養(yǎng),然后用碘液檢測(cè)淀粉水解圈的方式鑒定淀粉酶敲除情況,檢測(cè)結(jié)果見(jiàn)圖5,淀粉 酶基因敲除效率大于90%。由此可見(jiàn),本發(fā)明所得到的新的苦草芽孢具有很強(qiáng)的操作性,具 有廣泛的應(yīng)用前景。
