背景技術(shù):
葡萄球菌與醫(yī)院內(nèi)化膿性感染相關(guān),尤以金黃色葡萄球菌和表皮葡萄球菌最為密切,表皮葡萄球菌所致感染與一些高分子醫(yī)療器材在體內(nèi)的留置有關(guān),如靜脈導(dǎo)管、起搏器、腦脊液引流裝置、人工器官等。隨著上述醫(yī)療材料的使用日益頻繁,此類院內(nèi)感染問題日益嚴(yán)峻,且易發(fā)展為慢性感染,對抗生素的抗性較高。
已知在各種原核生物中普遍存在雙組分調(diào)控系統(tǒng),為原核細(xì)胞所特有的信號系統(tǒng),參與調(diào)控生物生長、毒力等多種生物學(xué)功能。通常由一對相應(yīng)的蛋白組成,稱為組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白。組氨酸蛋白激酶為跨膜蛋白,當(dāng)外界信號作用于組氨酸蛋白激酶的膜外區(qū)域,產(chǎn)生信號,使該蛋白發(fā)生自身磷酸化。隨后,磷酸化的組氨酸蛋白激酶作用于相應(yīng)的反應(yīng)調(diào)節(jié)蛋白發(fā)揮調(diào)控作用。不同的雙組分蛋白,調(diào)控細(xì)菌的不同生物學(xué)活性。然而,由于研究手段的局限性,對表皮葡萄球菌雙組分調(diào)控系統(tǒng)及對生物學(xué)功能的調(diào)控缺乏系統(tǒng)地了解和實(shí)際應(yīng)用。
發(fā)明內(nèi)容
本發(fā)明的目的是提供一組表皮葡萄球菌信號傳遞系統(tǒng)的新蛋白,包括組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白。本發(fā)明的另一目的是提供制備該類多肽的方法和編碼該類多肽的DNA,及其制備該類多肽的抗體,本發(fā)明的進(jìn)一步目的是利用該類多肽篩選藥物化合物。
本發(fā)明通過對表葡菌ATCC12228株的全基因組測序和序列分析,采用序列同源性比較和功能域分析等生物信息學(xué)方法確定了16對表皮葡萄球菌的雙組分調(diào)控信號蛋白(包括組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白),并制備了相應(yīng)的蛋白,經(jīng)對上述蛋白功能域空間結(jié)構(gòu)的模建和實(shí)驗(yàn)證實(shí)該類蛋白存在保守性結(jié)構(gòu)和活性位點(diǎn),可作為篩選藥物的“靶點(diǎn)”。
本發(fā)明表皮葡萄球菌組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白中包括16對雙組分調(diào)控蛋白,其中16個組氨酸蛋白激酶均具有結(jié)合ATP的激酶活性功能域(HATPase_c功能域),分別具有SEQ ID No.1-16的序列結(jié)構(gòu)。其中16個反應(yīng)調(diào)節(jié)蛋白均存在接受磷酸基團(tuán)的功能域(REC功能域),分別具有SEQ ID No.17-32的序列結(jié)構(gòu)。
本發(fā)明采用PMODELIHG軟件對上述相對保守的功能域片段進(jìn)行同源模建,結(jié)果顯示組氨酸蛋白激酶HATPase_c功能域的結(jié)構(gòu)特征是在AMP-PNP的結(jié)合部位形成袋狀結(jié)構(gòu),且在其周圍有保守的基序圍繞,所述AMP-PNP是一種ATP類似物;反應(yīng)調(diào)節(jié)蛋白的REC功能域模擬結(jié)構(gòu)由5個α螺旋和5個β折疊組成,其中存在三個天冬氨酸殘基構(gòu)成接受磷酸基團(tuán)的活性位點(diǎn),且接受磷酸基團(tuán)的過程需要二價陽離子(Mg2+或Mn2+)的輔助,所述天冬氨酸殘基是D7、D8、D51。
本發(fā)明所述的表皮葡萄球菌組氨酸蛋白激酶具有SEQ ID No.1-16的氨基酸序列結(jié)構(gòu),由SEQ ID No.33-48的DNA核苷酸序列編碼;所述的反應(yīng)調(diào)節(jié)蛋白具有SEQ ID No.17-32的氨基酸序列結(jié)構(gòu),由SEQ ID No.49-64的DNA核苷酸序列編碼。
本發(fā)明涉及由SEQ ID No.1-32構(gòu)成的蛋白的氨基酸序列的同源序列及片斷序列,和SEQ ID No.33-64編碼相關(guān)同源序列和片斷有關(guān)多肽的DNA核苷酸序列。所述同源DNA序列是指編碼SEQ ID No.33-64所示核苷酸序列或片斷,或DNA片斷長度在60個連續(xù)核苷酸區(qū)域內(nèi),一般至少70%同源于SEQ ID 1-32,可與之選擇性雜交的片斷核苷酸序列。所述SEQ ID 1-32的多肽同源序列至少在30個的連續(xù)氨基酸序列區(qū)域,至少60%以上的同源。
本發(fā)明的多肽包括組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白氨基酸序列的功能域部分序列,如具有生物學(xué)活性的氨基酸基本序列的多肽片斷;或其中部分氨基酸被性質(zhì)相似的氨基酸取代的多肽;或其氨基酸的一部分中加入或插入其他氨基酸的多肽,以及DNA的核苷酸序列改變,但不影響氨基酸序列的多肽,如同種氨基酸使用不同密碼子。
本發(fā)明提供了制備表皮葡萄球菌組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白(SEQ IDNo.1-32)方法,包括制備僅含生物學(xué)活性功能區(qū)的編碼序列的DNA片斷,克隆至適合的表達(dá)載體,轉(zhuǎn)染宿主菌,在適于的條件下培養(yǎng)該宿主細(xì)胞,以及從培養(yǎng)物中回收和純化所需蛋白。
上述技術(shù)方案中所有基本分子生物學(xué)操作均參照《分子克隆實(shí)驗(yàn)指南》。
本發(fā)明將編碼相應(yīng)蛋白的DNA序列與表達(dá)載體重組,形成重組表達(dá)質(zhì)粒。本發(fā)明不限于特定的表達(dá)質(zhì)粒。在一優(yōu)選實(shí)施方案中,本發(fā)明使用原核表達(dá)載體,例如pet28a等。
上述重組表達(dá)載體可按常規(guī)方法導(dǎo)入適宜宿主細(xì)菌。本發(fā)明不局限于任何特定的宿主細(xì)胞,只要它能夠表達(dá)所述重組表達(dá)載體。在一優(yōu)選實(shí)施方案中,本發(fā)明使用大腸桿菌JM109等。
本發(fā)明的多肽包括磷酸化反應(yīng)產(chǎn)物。
本發(fā)明還提供抗本發(fā)明多肽的多克隆抗體或單克隆抗體,使用本發(fā)明的多肽或其片斷作為免疫原,按常規(guī)方法制備特異性抗體??捎糜诩?xì)菌細(xì)胞內(nèi)的多肽表達(dá)量,也可用于研究所述多肽與細(xì)菌致病的關(guān)系,或用于阻斷細(xì)菌細(xì)胞之間的信號轉(zhuǎn)遞。
可以將本發(fā)明DNA的反義核酸的DNA插入合適的載體中,產(chǎn)生反義RNA;也可用合成方法產(chǎn)生反義RNA。反義RNA可用于控制細(xì)菌細(xì)胞內(nèi)多肽水平。
本發(fā)明多肽功能域結(jié)構(gòu)可用于篩選活性化合物,能阻斷組氨酸蛋白激酶或反應(yīng)調(diào)節(jié)蛋白質(zhì)間的磷酸基團(tuán)轉(zhuǎn)移和蛋白質(zhì)的磷酸化,抑制細(xì)菌細(xì)胞內(nèi)或細(xì)胞之間的信號傳遞,從而抑制葡萄球菌的生長。
本發(fā)明在表皮葡萄球菌ATCC12228株中確定了16對完整的雙組分調(diào)控系統(tǒng)的基因核苷酸序列及其功能。經(jīng)實(shí)驗(yàn)證實(shí)了組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白的磷酸化或兩者分子之間的磷酸基團(tuán)轉(zhuǎn)移可調(diào)控葡萄球菌的生物學(xué)活性,抑制組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白的磷酸化或磷酸基團(tuán)的轉(zhuǎn)移可抑制細(xì)菌的生長。對其保守性功能域的空間結(jié)構(gòu)模建確定了存在保守結(jié)構(gòu)和活性位點(diǎn)可作為篩選活性化合物的藥物“靶點(diǎn)”。
圖1是表皮葡萄球菌雙組分調(diào)控蛋白保守功能域空間結(jié)構(gòu)同源模建圖。其中,1a是組氨酸蛋白激酶HATPase_c功能域模擬結(jié)構(gòu),1b是反應(yīng)調(diào)節(jié)蛋白REC功能域模擬結(jié)構(gòu)。
具體實(shí)施例方式
實(shí)施例1 確定16對表皮葡萄球菌雙組分調(diào)控系統(tǒng)運(yùn)用序列同源性比較,功能域分析等生物信息學(xué)方法,在表皮葡萄球菌ATCC12228株全基因組(GeneBank收錄號AE015929)中確定了16對雙組分調(diào)控基因。表1列出了上述雙組分調(diào)控基因的基本情況,包括基因長度等,以及氨基酸序列同源性比較結(jié)果(blastp)。

通過與SMART氨基酸序列及功能域數(shù)據(jù)庫進(jìn)行同源性比較,對16對雙組分調(diào)控基因中存在的保守性功能域進(jìn)行分析,結(jié)果表明16個組氨酸蛋白激酶中均存在結(jié)合ATP的激酶活性功能域(HATPase_c功能域),16個反應(yīng)調(diào)節(jié)蛋白(SEQID No.17-32)均存在接受磷酸基團(tuán)的功能域(REC功能域),表2是表皮葡萄球菌雙組分調(diào)控基因功能域分析結(jié)果。

實(shí)施例2 對HATPase_c功能域和REC功能域進(jìn)行空間結(jié)構(gòu)模建根據(jù)上述確定的HATPase_c功能域和REC功能域在雙組分調(diào)控系統(tǒng)中的相對保守性,運(yùn)用PMODELING軟件對兩個功能域進(jìn)行空間結(jié)構(gòu)的同源模建。結(jié)果顯示HATPase_c功能域模擬結(jié)構(gòu),與已報道的大腸桿菌組氨酸蛋白激酶基因EnvZ的HATPase_c功能域的和磁共振(NMR)結(jié)構(gòu)非常相似(Ncbi mmdbId11091),即HATPase_c功能域主要結(jié)構(gòu)由4個β折疊和3個α螺旋組成,尤其是在AMP-PNP(一種ATP類似物)的結(jié)合部位形成一個袋狀結(jié)構(gòu),且其周圍有一些保守的基序(G1 box、G2 box、F box)圍繞,見圖1a;而REC功能域模擬結(jié)構(gòu)與大腸桿菌的反應(yīng)調(diào)節(jié)蛋白基因CheY的REC功能域的和磁共振(NMR)結(jié)構(gòu)也非常相似(NcbimmdbId3000),由5個α螺旋和5個β折疊組成,其中存在三個天冬氨酸殘基(D7、D8、D51)構(gòu)成接受磷酸基團(tuán)的活性位點(diǎn),且該過程需要二價陽離子(Mg2+或Mn2+)的輔助,見圖1b。因此,這兩個結(jié)構(gòu)均存在藥物設(shè)計、篩選的“靶標(biāo)”,以它們?yōu)槟0澹Y選各類小分子化合物庫,能找到一些在空間結(jié)構(gòu)上封閉該活性結(jié)構(gòu)或活性位點(diǎn)的小分子化合物,從而阻斷這些雙組分調(diào)控系統(tǒng),其作用原理是阻斷雙組分調(diào)控系統(tǒng)信號傳導(dǎo)途徑的一個或多個環(huán)節(jié),包括組氨酸蛋白激酶的自身磷酸化、將磷酸基團(tuán)從蛋白激酶轉(zhuǎn)移到調(diào)節(jié)蛋白的轉(zhuǎn)磷酸基過程以及反應(yīng)調(diào)節(jié)蛋白的磷酸化等。
實(shí)施例3 16對雙組分調(diào)控基因相關(guān)功能的分析將16對雙組分調(diào)控基因與已報道的其他革蘭陽性菌中有明確功能的雙組分基因進(jìn)行了序列同源性比較,結(jié)果發(fā)現(xiàn)有8對雙組分調(diào)控基因與已報道的其他革蘭陽性菌中雙組分基因有較高的氨基酸序列同源性(同源性大于30%),表明所述雙組分調(diào)控系統(tǒng)在同為革蘭陽性菌的表皮葡萄球菌中也可能發(fā)揮著重要的作<p>表2-(4周時的結(jié)果)
將上述重組質(zhì)粒1175-pet28a、1176-pet28a、1175d-pet28a按常規(guī)方法在IPTG誘導(dǎo)下表達(dá),用SDS-PAGE電泳對目的蛋白進(jìn)行鑒定,結(jié)果顯示三種重組質(zhì)粒分別表達(dá)70kd、30kd及30kd左右的蛋白,用申能博彩公司提供的3s NTAHis-Tag Resin層析柱進(jìn)行蛋白提純。
實(shí)施例5 其余1 5對雙組分基因(除外se1175/se1176)原核表達(dá)質(zhì)粒的構(gòu)建以表葡菌ATCC12228株全基因組DNA為模板,設(shè)計相應(yīng)引物經(jīng)PCR獲得。

4.根據(jù)權(quán)利要求1所述的表皮葡萄球菌調(diào)節(jié)信號多肽,其特征在于所述的組氨酸蛋白激酶的SEQ ID No.1-16的氨基酸序列結(jié)構(gòu),由SEQ ID No.33-48的DNA核苷酸序列編碼;所述的反應(yīng)調(diào)節(jié)蛋白的SEQ ID No.17-32的氨基酸序列結(jié)構(gòu),由SEQ ID No.49-64的DNA核苷酸序列編碼。
5.根據(jù)權(quán)利要求1所述的表皮葡萄球菌調(diào)節(jié)信號多肽,其特征在于還具有由SEQID No.1-32構(gòu)成的蛋白的氨基酸序列的同源序列及片斷序列,和SEQ ID No.33-64編碼相關(guān)同源序列和片斷有關(guān)多肽的DNA核苷酸序列。
6.根據(jù)權(quán)利要求5所述的表皮葡萄球菌調(diào)節(jié)信號多肽,其特征在于所述的同源DNA序列是編碼SEQ ID No.33-64所示核苷酸序列或片斷,或DNA片斷長度在60個連續(xù)核苷酸區(qū)域內(nèi),至少70%同源或可與之選擇性雜交的片斷核苷酸序列;所述SEQ ID 1-32的多肽同源序列至少在30個連續(xù)氨基酸序列區(qū)域,至少60%以上的同源。
7.基于權(quán)利要求1所述的多肽,其多肽的磷酸化反應(yīng)產(chǎn)物。

本發(fā)明屬生物技術(shù)領(lǐng)域,涉及作為表皮葡萄球菌信號傳遞系統(tǒng)關(guān)鍵新蛋白組氨酸蛋白激酶和反應(yīng)調(diào)節(jié)蛋白;制備該類多肽的方法;編碼該類多肽的DNA;該類多肽的抗體;以及利用該類多肽結(jié)構(gòu)功能域篩選藥物化合物。本發(fā)明篩選的活性化合物,能阻斷組氨酸蛋白激酶或反應(yīng)調(diào)節(jié)蛋白質(zhì)間的磷酸基團(tuán)轉(zhuǎn)移和蛋白質(zhì)的磷酸化,抑制細(xì)菌細(xì)胞內(nèi)或細(xì)胞之間的信號傳遞,從而抑制葡萄球菌的生長。
