本發(fā)明涉及一種9,10-開環(huán)留體類化合物及其制備方法與應用。
背景技術:
自20世紀60年代從柳珊瑚中發(fā)現(xiàn)了具有重要生理活性的前列腺素開始,國內(nèi)外許多研究組已經(jīng)從柳珊瑚中發(fā)現(xiàn)了大量的抗腫瘤化合物,如萜類、留體、生物堿和脂類等結構類型,但是尚未開發(fā)出有效的抗腫瘤藥物。9,10-開環(huán)留體類化合物是一類重要的海洋次級代謝產(chǎn)物,目前已從柳珊瑚Astrogorgia sp. ,Calicogorgia sp.和 Muricella sp.中分離獲得此類化合物 12 個。該類化合物具有強抗菌、細胞毒、鹵蟲致死等生物活性。但到目前為止,國內(nèi)外均未見對軟柳 珊瑚Subergorgia rubra次級代謝產(chǎn)物的報道,也未見關于含有8-輕基的9,10-開環(huán)甾體類化合物結構及活性報道。
發(fā)明內(nèi)容
本發(fā)明的目的在于提供一種來源于軟柳珊瑚S. rubra的9,10-開環(huán)甾體類化合物及其制備方法與作為抗腫瘤藥物的應用,它能滿足現(xiàn)有技術的上述需求。一種9,10-開環(huán)甾體類化合物,其特征在于它的結構式為

II O上述9,10-開環(huán)甾體類化合物的制備方法,其特征在于將軟柳珊瑚S. rubra用乙醇提取,提取液減壓濃縮得到浸膏,將浸膏進行減壓硅膠柱層析,用石油醚-乙酸乙酯梯度洗脫,再經(jīng)S^hadex LH-20凝膠柱層析及制備高效液相色譜等分離方法得到化合物Subergorgiaols A M0上述的9,10-開環(huán)甾體類化合物在制備抗腫瘤劑中的應用。本發(fā)明從軟柳珊瑚S. rubra中獲得新的9,10-開環(huán)甾體類化合物對宮頸癌細胞caski和肝癌細胞IfepG-2具有強的抑制活性,可用作抗腫瘤藥物,應用前景廣闊。
具體實施方式
本發(fā)明的化合物的制備(I)取新鮮軟柳珊瑚S. rubra樣品(1800. Og),粉碎后用95% (體積百分數(shù))乙醇水溶液浸提3次,合并提取液,減壓濃縮得粗浸膏(38. 本發(fā)明涉及一種9,10-開環(huán)甾體類化合物及其制備方法與應用,制備時先用乙醇提取軟柳珊瑚Subergorgia rubra,將提取液減壓濃縮得到浸膏,將該浸膏用硅膠柱層析、凝膠柱層析和高效液相色譜進行分離,得到化合物Subergorgiaols A~M。本發(fā)明來源于珊瑚的9,10-開環(huán)甾體類化合物對宮頸癌細胞caski和肝癌細胞HepG-2具有強的抑制活性,可用于制備抗腫瘤藥物,應用前景廣闊。
