本發(fā)明涉及三萜類化合物,具體涉及從真菌中提取的三萜類化合物及其作為醛糖還原酶抑制劑的用途。
背景技術(shù):
:靈芝(ganodermalucidum(leyss.exfr.)karst.)隸屬于真菌門(eumycota)、擔子菌亞門(basidiomycotina)、層菌綱(hymenomycetes)、非褶菌目(aphyllophorales)、靈芝科(gamodermataceae)、靈芝屬(ganoderma)類?!吨腥A本草》中記錄“其性甘;味平;無毒;歸肺、心、脾、腎經(jīng)。益氣血;安心神;健脾胃。主治虛勞、心悸、失眠、頭暈、神疲乏力、久咳氣喘、冠心病、矽肺、腫瘤?!薄吨袊幍洹分袑㈧`芝收錄為中藥材,其中的靈芝多糖、三萜、核苷、生物堿、氨基酸多肽、微量元素等成分是其藥效物質(zhì)的主要基礎(chǔ),以三萜和多糖為其中主要活性成分?,F(xiàn)代藥理研究表明,靈芝三萜類化合物具有保肝、抗腫瘤、抗hiv-4及hiv-4蛋白酶活性、抗組織胺釋放、抑制血管緊張素、抗氧化等作用。代謝綜合征主要表現(xiàn)為胰島素抵抗,中心肥胖,提高血壓和血脂,并已經(jīng)成為世界性的疾病。目前雖然有一些臨床治療代謝綜合征的藥物比如胰島素、他汀類以及貝特類藥物,但是由于其副作用以及治療功效不夠理想等仍然是這類治療藥物面臨很多問題。醛糖還原酶(aldosereductase,ar)存在于人體神經(jīng)、紅細胞、晶狀體、視網(wǎng)膜等組織器官中,在多元醇通路中催化血液中的葡萄糖生成山梨醇,是多元醇通路的關(guān)鍵限速酶。當血糖濃度維持在正常生理水平時,它并不激活,對葡萄糖的親和力較低,此時葡萄糖很少轉(zhuǎn)化為山梨醇。在高血糖狀況下(如糖尿病),己糖激酶被飽和,這時醛糖還原酶激活,促使體內(nèi)的葡萄糖轉(zhuǎn)化為山梨醇。然而,山梨醇脫氫酶(sorbitoldehydrogenase,sdh)的活力并未相應地成比例增加,山梨醇轉(zhuǎn)化為果糖的效率沒有提高。山梨醇本身由于極性強不易通過細胞膜,在細胞內(nèi)會形成蓄積,使細胞膜的通透性發(fā)生改變,并使細胞中na+-k+-atp酶活性下降,造成肌醇喪失,導致細胞代謝與功能的損害。由于眼睛和神經(jīng)細胞等組織內(nèi)醛糖還原酶的含量較高,糖尿病病人體內(nèi)高血糖的環(huán)境使這一通路很容易被打開,造成對這些組織的病理損害,如糖尿病、白內(nèi)障、神經(jīng)病變、腎臟病變、視網(wǎng)膜病變、動脈粥樣硬化等糖尿病并發(fā)癥(diabetescontrolandcomplications,dcc)糖尿病并發(fā)癥嚴重威脅著糖尿病人的生存質(zhì)量,阻斷或減弱醛糖還原酶活性的藥物可以用來預防或推遲糖尿病并發(fā)癥的發(fā)生。1970年代以來,醛糖還原酶抑制劑的研究成為治療糖尿病藥物研究的熱點,已有一些種類進入臨床試驗或上市銷售,但是好多藥物有已經(jīng)由于臨床試驗存在問題而退出。目前可供臨床使用的醛糖還原酶抑制劑依然很少在我們的長期研究中,從峨眉山赤芝的子實體乙酸乙酯粗提物中分離到具有醛糖還原酶抑制活性的化合物。這些化合物表現(xiàn)出了和陽性藥依帕司他相似的活性,為醛糖還原酶抑制劑藥物開發(fā)提供先導化合物。技術(shù)實現(xiàn)要素:本發(fā)明以靈芝為研究對象,對其提取物進行了分離純化,得到新的三萜以及雜萜類化合物,對這些化合物進行了結(jié)構(gòu)鑒定和生物活性研究,發(fā)現(xiàn)這些化合物具有很好的醛糖還原酶抑制活性,有潛在的藥用價值。為此,本發(fā)明提供一種通式(i)所示的化合物或其藥用鹽:其中,碳8和碳9之間為雙鍵,或者碳7與碳8之間和碳9與碳11之間為雙鍵;r1為=o,α-oh或β-oh,r2為-h或-oh,r3為h、=o或β-oh,r4為-h或=o,r5為-h、β-oh或β-ococh3,r6為-h、α-oh、β-oh、=o或α-ococh3,r7為-ch(ch3)ch2c(o)ch2ch(ch3)cooch3、-ch(ch3)ch2c(o)ch2ch(ch3)cooh、-ch(ch3)ch2ch(oh)ch=c(ch3)cooh、-(ch3)c=chc(o)ch2ch(ch3)cooch3、-ch(ch3)ch2ch2ch(oh)c(ch3)(oh)ch3、-ch(ch3)ch2ch2ch=c(ch2oh)cooh、-ch(ch3)ch2ch2ch=c(ch3)cooh、-ch(ch3)ch2ch2ch(r9)c(ch2r8)(oh)ch3,r8為-oh、r9為α-oh或優(yōu)選的,化合物為如下式(i-1)到(i-7)的化合物:r1r2r3r4r5r61β-ohoh=oβ-ococh3=och32β-ohhβ-ohhα-ohh3β-ohhβ-ohhβ-ohch34=ohβ-ohh=och35=oh=oβ-ococh3=ohr1r2r36=ococh3α-oh7β-ohhβ-ohr1r2r3r410=ohch2ohcooh11α-ohα-ococh3coohch312=oα-ococh3coohch3r1r213α-ohc14α-oha15β-aoh16α-ohb化合物1-17的結(jié)構(gòu)式如下:具體實施方式下述實施例中所使用的實驗方法如無特殊說明,均為常規(guī)方法。下述實施例中所用的材料、試劑等,如無特殊說明,均可從商業(yè)途徑得到。hplc分析儀為agilent1200分析型液相色譜儀。hplc的條件如下:以色譜甲醇將樣品配制為10mg/ml的溶液,上樣量為每次15ul,色譜柱為kromasil10×250mmc18半制備柱,柱溫為25℃,210nm波長進行hplc制備。實施例1制備靈芝的提取物將靈芝子實體剪碎,稱重2500克。用10l體積百分含量95%乙醇的水溶液回流提取3次,每次1小時。合并提取液,減壓濃縮干燥得到170克提取物。進一步將提取物用蒸餾水600ml溶解,用等體積的正己烷萃取三次,棄去有機相。再用等體積乙酸乙酯萃取水相三次,棄去水相,合并乙酸乙酯萃取液。用旋轉(zhuǎn)蒸發(fā)儀蒸干乙酸乙酯獲得浸膏49g,記作gs。實施例2化合物的制備靈芝粗提物乙酸乙酯層gs通過硅膠柱色譜分離,以正己烷:乙酸乙酯體系(正己烷:乙酸乙酯的體積比為1:0,50:1,30:1,20:1,15:1,10:1,4:1,2:1),二氯甲烷:甲醇體系(二氯甲烷:甲醇的體積比為1:0,100:1,50:1,30:1,10:1,5:1)依次進行梯度洗脫,每個梯度洗滌4個保留體積,每個保留體積一個餾分。根據(jù)薄層層析色譜行為分析,合并相似流分,在正己烷:乙酸乙酯(體積比為1:0)體系得到餾分gs-1;在正己烷:乙酸乙酯(體積比為100:1~30:1)中得到餾分gs-2~5;在正己烷:乙酸乙酯(體積比為20:1)中得到餾分gs-6,gs-7;在正己烷:乙酸乙酯(體積比為10:1)中得到餾分gs-8;在二氯甲烷:甲醇(體積比為1:0)中得到餾分gs-9~10;在二氯甲烷:甲醇(體積比為50:1)中得到餾分gs-11~13;在二氯甲烷:甲醇(體積比為20:1)中得到餾分gs-14在二氯甲烷:甲醇(體積比為30:1)中得到餾分gs-15;在二氯甲烷:甲醇(體積比為20:1,10:1,1:0)中得到餾分gs-16。共得到16個餾分,將各餾分冷凍干燥。對gs-7利用ods反相硅膠柱進一步分離。用體積百分含量為10%,20%,30%,40%,50%,70%,90%的甲醇水溶液依次進行洗脫,每個洗脫體系洗3個保留體積,每個保留體積為500ml。根據(jù)薄層層析色譜行為,照射254nm紫外燈判斷不同餾分,在體積百分含量為20%~40%的甲醇水溶液中得到餾分gs-7-1~3;在體積百分含量為50%的甲醇水溶液系統(tǒng)中得到餾分gs-7-4;在體積百分含量為60%的甲醇水溶液系統(tǒng)中得到餾分gs-7-5~6;在體積百分含量為70%,90%的甲醇水溶液系統(tǒng)中得到餾分gs-7-7;共得到7個子餾分,將各個餾分冷凍干燥。對gs-7-3餾分進一步進行hplc制備。以體積百分含量40%乙腈的酸水(此處的酸水為體積百分含量為0.01%三氟乙酸的水溶液)溶液為洗脫劑進行hplc制備,流速為2ml/min,收集25min的色譜峰,得到化合物1;收集28min的色譜峰,得到化合物2;收集31min,35min的色譜峰,分別得到化合物3、4。對gs-7-4以體積百分含量40%乙腈的酸水(此處的酸水為體積百分含量為0.01%三氟乙酸的水溶液)溶液為洗脫劑進行hplc制備,流速為2ml/min,收集20min的色譜峰,得到化合物5;收集23.4min的色譜峰,得到化合物6;收集28和30min的色譜峰,分別得到化合物7、8。對gs-7-5以體積百分含量42%乙腈酸水(此處的酸水為體積百分含量為0.01%三氟乙酸的水溶液)溶液為洗脫劑進行hplc制備,流速為2ml/min,分別收集23,27.2min的色譜峰得到化合物9、10。對gs-7-4以體積百分含量45%乙腈的酸水(此處的酸水為體積百分含量為0.01%三氟乙酸的水溶液)溶液為洗脫劑進行hplc制備,流速為2ml/min,收集23.2min的色譜峰,得到化合物11;收集27.3min的色譜峰,得到化合物12。通過薄層色譜分析還發(fā)現(xiàn)gs-9中活性物質(zhì)含量較高,因此對gs-9利用ods反相硅膠柱分離。用體積百分含量為20%,40%,50%,70%,90%甲醇的水溶液依次進行洗脫,每個洗脫體系洗1500ml。根據(jù)薄層層析色譜行為,照射254nm紫外燈判斷不同餾分,在體積百分含量為20%的甲醇水溶液中得到餾分gs-9-1~3;在體積百分含量為40%的甲醇水溶液系統(tǒng)中得到餾分gs-9-4~6;在體積百分含量為50%的甲醇水溶液系統(tǒng)中得到餾分gs-9-7~8;在體積百分含量為70%的甲醇水溶液系統(tǒng)中得到餾分gs-9-9~10;在體積百分含量為90%的甲醇水溶液系統(tǒng)中得到餾分gs-9-11。共得到11個子餾分,將各個餾分冷凍干燥。通過薄層色譜分析發(fā)現(xiàn)gs-9-6中活性物質(zhì)含量較高,因此對gs-9-6進一步進行hplc分離制備。以體積百分含量82%甲醇的酸水(此處的酸水為體積百分含量為0.01%三氟乙酸的水溶液)溶液為洗脫劑進行hplc制備,流速為2ml/min,收集15min的色譜峰,得到化合物13;收集19min的色譜峰,得到化合物14;收集23min,27min的色譜峰,分別得到化合物15、16;收集30min的色譜峰,分別得到化合物17。實施例3化合物1-17的確認對制備得到的17個化合物分別進行核磁共振、紅外、質(zhì)譜檢測,確定各化合物的結(jié)構(gòu)。其中所使用的核磁共振儀為varianmercury-500和-600兆赫,紅外色譜儀為nicoletis5ft-ir,質(zhì)譜儀為brukerapexiii7.0t和apexiift-icr。經(jīng)確認,化合物1、6、8、10、13、14、15為新化合物,均含有三萜骨架i。1、6、8、10、13、14、15的nmr的碳譜和氫譜確認數(shù)據(jù)如表1-2所示?;衔?(lucidenicacidr)化合物1結(jié)構(gòu)如下:碳8和碳9之間為雙鍵r1為β-oh,r2為-oh,r3為=o,r4為=o,r5為β-ococh3,r6為=o,r7為-ch(ch3)ch2c(o)ch2ch(ch3)cooh,(6r)-6-((3s,4r,5r,10s,12s,13r,14r,17r)-12-乙酰氧基-3-羥基-4-(羥基甲基)-4,10,13,14-四甲基-7,11,15-三氧代-2,3,4,5,6,7,10,11,12,13,14,
